چکیده

Abstract Hypertrophic scar (HS) is a kind of serious pathological with no currently effective treatment. HS fibroblasts (HSFs) are the main effector cells for formation. Ellagic acid (EA) exerts regulatory effects in some diseases, but its role remains unclear. This study aimed to evaluate effect EA on fibrotic phenotypes HSFs and further investigate downstream signaling mechanism. The cell counting kit-8 (CCK-8) assay was used perform cytotoxicity proliferation assays. migration assessed using wound healing transwell contraction measured by collagen lattice detection ?-smooth muscle actin (?-SMA) expression. levels mRNA protein were determined qPCR western blotting, respectively. results showed that inhibited proliferation, migration, expression dose-dependent manner. Furthermore, not only suppressed Smad2/3 pathway also reversed TGF-?1-induced activation up-regulation cellular HSFs. These findings demonstrate anti-fibrotic blocking TGF-?1/Smad2/3 pathway, which indicates potential therapeutic candidate treatment HS.

Title Ellagic acid, a phenolic compound, exerts anti-angiogenesis effects via VEGFR-2 signaling pathway in breast cancer

Anti-angiogenesis targeting VEGFR-2 has been considered as an important strategy for cancer therapy. Ellagic acid is a naturally existing polyphenol widely found in fruits and vegetables. It was reported that ellagic acid interfered with some angiogenesis-dependent pathologies. Yet the mechanisms involved were not fully understood. Thus, we analyzed its anti-angiogenesis effects and mechanisms ...

Ellagic Acid Exerts Anti-proliferation Effects on MCF-7 Breast Cancer Cells

Ellagic acid is a naturally occurring polyphenolic compound present in pomegranate plants, strawberries, grapes, apples, tendril blueberries, blackberries and walnuts. Ellagic acid exhibits a variety of effects such as anti-bacterial, anti-inflammatory, immune regulatory and anti-tumor effects (Chen et al., 2011). In particular, ellagic acid has an inhibitory effect on many tumors, and is curre...

Gamma Linolenic Acid Exerts Anti-Inflammatory and Anti-Fibrotic Effects in Diabetic Nephropathy

PURPOSE This study was undertaken to investigate the effects of gamma linolenic acid (GLA) on inflammation and extracellular matrix (ECM) synthesis in mesangial and tubular epithelial cells under diabetic conditions. MATERIALS AND METHODS Sprague-Dawley rats were intraperitoneally injected with either a diluent [n=16, control (C)] or streptozotocin [n=16, diabetes (DM)], and eight rats each f...

MicroRNA-21 Regulates hTERT via PTEN in Hypertrophic Scar Fibroblasts

BACKGROUND As an important oncogenic miRNA, microRNA-21 (miR-21) is associated with various malignant diseases. However, the precise biological function of miR-21 and its molecular mechanism in hypertrophic scar fibroblast cells has not been fully elucidated. METHODOLOGY/PRINCIPAL FINDINGS Quantitative Real-Time PCR (qRT-PCR) analysis revealed significant upregulation of miR-21 in hypertrophi...

Effects of TGF-beta and TGF-beta-neutralizing antibody on normal skin fibroblasts and scar-derived fibroblasts.

Transforming growth factor-beta (TGF-beta) is known as an important cytokine for scar formation in wound healing. The purpose of the present study was to investigate the effects of TGF-beta and its neutralizing antibody on normal skin fibroblasts and scar-derived fibroblasts in culture. Endogenous TGF-beta levels were similar in all fibroblasts. Cell proliferation increased when TGF-beta 1 or b...
